Association of Alpha-Methylacyl-CoA Racemase Gene Polymorphisms with Susceptibility to Prostate Cancer: a Meta-Analysis

چکیده
Prostate cancer (PCa) is one of the most common malignancies in men with approximately 0.9 million newly diagnosed cases and 0.26 million deaths in 2008 worldwide (Jemal et al., 2011). Until now, the exact carcinogenic mechanism of prostate cancer has remained unclear (Mahmoud et al., 2014). It is widely accepted that carcinogenesis of prostate cancer is affected by genetic susceptibility and environmental exposure, for instance, a fatty acid diet (Amankwah et al., 2012; Doolan et al., 2014; Askari et al., 2014). Single nucleotide polymorphisms (SNPs), as a type of genetic predisposing factor, might lead to changes in protein encoding and be involved in the alterative risk of prostate cancer (Yang et al., 2013; Zhang et al., 2014). Therefore, it is crucial to focus on these genetic markers for their application in susceptibility prediction, early diagnosis and individualized therapeutic regimens of prostate cancer.

SequenceVariation ina-Methylacyl-CoARacemase andRiskof Early-Onset and Familial ProstateCancer
BACKGROUND. Expression of the a-methylacyl-CoA racemase (AMACR) gene has been established as a sensitive and specific biomarker for the diagnosis of prostate cancer. An initial study has also suggested that the risk of familial (but not sporadic) prostate cancer may be associated with germline variation in the AMACR gene. METHODS. In a study of brothers discordant for the diagnosis of prostate ...
متن کاملDecreased alpha-methylacyl CoA racemase expression in localized prostate cancer is associated with an increased rate of biochemical recurrence and cancer-specific death.
Alpha-methylacyl CoA racemase (AMACR) is overexpressed in prostate cancer relative to benign prostatic tissue. AMACR expression is highest in localized prostate cancer and decreases in metastatic prostate cancer. Herein, we explored the use of AMACR as a biomarker for aggressive prostate cancer. AMACR protein expression was determined by immunohistochemistry using an image analysis system on tw...
متن کاملA-Methylacyl-CoA Racemase (AMACR) and Prostate-Cancer Risk: A Meta-Analysis of 4,385 Participants
BACKGROUND Alpha-methylacyl-CoA racemase (AMACR) is a mitochondrial and peroxisomal enzyme that is overexpressed in prostate cancer. The aim of this study was to confirm and expand the findings that the PCa risk increased in men associated with AMACR expression across various geographic regions. METHODS A systematic search of databases was carried out and other relevant articles were also ide...
متن کاملEvaluation of alpha-methylacyl-CoA racemase, metallothionein and prostate specific antigen as prostate cancer prognostic markers.
Current diagnostic techniques are inefficient in distinguishing latent and low-risk forms of prostate cancer from high-risk forms. The present study is focused on determination of putative tumor markers of aggressive high-grade forms of prostate cancer. Potential markers were determined in blood sera of 133 patients (82 cases and 51 controls) and in cell lines (Gleason score 9-derived 22Rv1 and...
متن کاملGenetic Variations of α-Methylacyl-CoA Racemase Are Associated with Sporadic Prostate Cancer Risk in Ethnically Homogenous Koreans
BACKGROUND To assess if the variants of (R)-alpha-methyl-CoA racemase (AMACR) gene would be associated with the risk of sporadic prostate cancer in ethnically homogenous Koreans. MATERIALS AND METHODS We enrolled 194 patients with prostate cancer and 169 healthy controls. A total of 17 single nucleotide polymorphisms of the AMACR gene were selected. The distribution of each genotype and haplo...
